Not sure how feasible this is but when I respond to calendar invites that came from an addy.io alias, it sends the response from the forwarded address rather than the alias.

Is there a way to modify the calendar invite itself so that responses are handled correctly?

I don't believe this is specific to any given email client as all have some sort of "accept" button, but yes, I am talking about having some way of responding yes to an event invitation using an alias. Ideally using any client's native button.
